Abstract
This document specifies a method for the determination of the estrogenic potential of water and wastewater using a reporter gene assay with genetically modified yeast strains of Saccharomyces cerevisiae. This reporter gene test is based on the activation of the human estrogen receptor alpha. The p-YES method enables an unambiguous fingerprint of a sample due to the previous separation and fractionation of the estrogen-active components of a sample according to their polarity and the subsequent effect-related detection. With this method, the concentrations of E1, EE2, E2 and E3 can be determined and the respective effect equivalents of all other estrogen-active components can be determined. This method is suitable with a high sensitivity for extracts from the following water samples: - Treated and untreated wastewaters; - leachates; - eluates and pore water of sediments; - Single substances and mixtures of substances; - surface water; - groundwater; - raw and drinking water. The limit of quantification (LOQ) of this method for the analysis of water samples is between 0,5 pg and 1,0 pg for E2 and EE2 in the test.
